In tempo di pace operò sulla costa orientale, mentre, al momento di Pearl Harbour si trovava nei cantieri di Norfolk. Trasferita immediatamente nel Pacifico, all'inizio del 1942 operò al largo della Nuova Guinea, attaccando naviglio giapponese e instalazioni costiere. Raggiunta a maggio dalla Lexington, attaccò la portaerei leggera giapponese Soho nel Mar dei Coralli, affondandola con i propri aerei e da#HISTORY OF THE CARRIER YORKTOWN CV5nneggiando gravemente la portaerei pesante Shokaku. 

La Yorktown fu gravemente danneggiata (tanto che i giapponesi ritenevano di averla affondata!) ma fu rimessa in grado di combattere a tempo di record. Partecipò attivamente alla battaglia di Midway. Ingaggiò un epico duello con la portaerei giapponese Soryu, affondandola e venendo essa stessa colpita a morte da due ondate di bombardieri e areosiluranti provenienti dalla Soryu stessa. 

Rimase immobilizzata e due giorni dopo, mentre era rimorchiata verso Pearl Harbour fu raggiunta dai siluri del sommergibile giapponese I-168; il giorno dopo affondò.

In time of peace it operated on the oriental coast, while, during Pearl Harbour it was found in the yards of Norfolk. Immediately moved to the Pacific, to the beginning of 1942 it operated offshore the New Guinea attaching Japanese shipping and coastal instalazioni. Reached in May by the Lexington, it attached the Japanese light aircraft carrier Soho in the Sea of the Corals, sinking it with its own airplanes and seriously damaging the heavy aircraft carrier Shokaku. 

The Yorktown was seriously damaged (so much that the Japanese held to have sunk it!) but able was put again to fight to time of record. It actively participated in the battle of Midway. It engaged an epic duel with the Japanese aircraft carrier Soryu, sinking it and coming it same stricken to died by two waves of bombardiers and areosiluranti coming from the Soryu stessa. 

It was tied-up and two days later, while it was being towed toward Pearl Harbour it was reached by the torpedos of the Japanese submersible The-168; later the day sank.
